Swap out 3rz for a 2rz?

My friend has a 2rz that he wants to put in my 1999 tacoma 2.7L 4x4. Is it going to change anything? will 4x4 still work?

What exactly is your friend proposing to do?

Transplant a whole 2RZ "as is" to replace your 3RZ?

Or ...

Use a 2RZ block to build up a new 3RZ using the 3RZ head and internals?

You will notice the loss of torque.
Not sure why you would ask if 4x4 would still work... are you planning on replacing transmission as well?
